Overview
This configuration allows shorter cycle times by limiting the acquisition time to just the peaks of interest and regenerating the alternating column during the sample acquisition[1]. It uses
Cycle Composer Software[2] - this application is run with Standard LEAP macros outside of Analyst® Software[3] from Applied Biosystems Inc®
If the peaks of interest come off together in the middle of the gradient (as shown below) then it is possible to utilize the front cut [4] and back cut [5] time to increase throughput.
This application is useful to increase sample throughput by taking advantage of front and/or back-cut time to load the next sample in the sequence early. It requires that the "period of interest" in the chromatography is significantly shorter than the overall chromatographic run-time. There are two columns that are running in "staggered" mode[6], where samples are pre-loaded onto the other channel while the previous channel is still in acquisition. There is no pre-column. The gradient programs regenerate [7] the analytical columns in each stream.
Please note: LEAP can not control the MS Selector valve as part of the cycle.
If injections are staggered in the following way, there is a potential for up to 100% gain:
